INFLUENCE OF THE USE OF REFRIGERATED SEMEN ON THE IN VITRO PRODUCTION OF BUBALINE EMBRYO

SUMMARY:

Semen is one of the factors that most interfere with the in vitro production of buffalo embryos, therefore, the aim of this study will be to evaluate the influence of chilled and frozen semen on embryo production. Four Murrah bulls will be used, which will be submitted to clinical-andrological examination and subsequently submitted to semen collection by the artificial vagina method, 24 hours before in vitro fertilization. The semen will be evaluated for its physical, morphological and kinetic characteristics, through optical microscopy and computerized evaluation (CASA). Subsequently, the semen will be divided into aliquots and separated into two groups (G1 - frozen and G2 - refrigerated). After processing the semen will be used in the production of in vitro embryos. Oocytes will be collected in vivo by ultrasound-guided follicular aspiration (OPU), classified and placed in maturation drops for a period of 20 hours. The matured oocytes will be fertilized with semen from G1 and G2. After 72 hours of fertilization, the zygotes will be evaluated for cleavage and kept in culture drops for 3 days until the blastocyst stage. The data will be evaluated regarding the difference in the embryo production rate of the groups through electronic spreadsheets and submitted to statistical analysis in specialized software (SAS 2022). The chi-square statistical test will be used with a significance level of 5%.
