The immunocastration procedure has been used as a technical alternative largely favorable to animal welfare, as it is painless, less invasive and with efficacy similar to that of surgical castration. Therefore, the objective was to analyze the possible effects of immunocastration on the buffalo's testicular parenchyma. Twenty buffalo bulls, aged between two and three years, from the island of Marajó - PA, were randomly divided into two groups (n = 10 in each group): control group (CG) immunocastrated group (GIM). The product used was the anti-GnRH Bopriva® vaccine (Zoetis, SP, Brazil). The GIM (n = 10) received two doses of 1.0ml containing 400μg of the anti-GnRH Bopriva® vaccine with an interval of eight weeks between applications and the control group received 1.0 ml of saline. Fourteen days after the last dose, the twenty animals were slaughtered (and the testicles were immediately collected and dissected and then measured (length, width, weight and circumference) and fragments of the testicular parenchyma were removed for making slides and later histological analysis. Significant differences (p> 0.05) were observed in length, width, circumference and weight .The weight of the right and left testicles of the immunocastrated group decreased by approximately 54%, as well as the bilateral circumference showed a reduction of approximately 37%, the bilateral width reduced approximately 14% and the length had a significant reduction of approximately 9%, with significant changes in the size of the seminiferous tubule area of 15%, concomitantly with the reduction, in 48% in the area of the lumen. testicular presented a moderate to severe degeneration. The main alterations were: desquamation (degeneration seminiferous epithelium, presence of vacuolization of Sertoli cells, reduction in the number of Leydig cells, pronounced intertubular fibrosis, pycnotic nucleus and azoospermia in 90 to 100% of the animals. We conclude that the anti-GnRH vaccine was effective in causing lesions in the testicular parenchyma, significantly compromising spermatogenesis probably caused by decreased testosterone production
