SUMMARY:

The environmental changes resulting from human actions have caused the appearance of high concentrations of heavy metals, introduced into the environment by the intensification of fertilizers, agrochemicals, mining and metallurgy. Cadmium (Cd) is a bioaccumulator and can cause renal failure in mammals in plants enzymatic degradation. To reduce and cancel the toxic effects present in polluted areas, many environmental restoration projects have been carried out with the adoption of plant species, which have physiological mechanisms capable of extracting and solubilizing harmful chemical elements. Phytoremediation offers management of contaminated regions in order to reduce costs and enhance the reuse of degraded territory using plants of low economic value and high ecological importance. Thus, the present study aims to analyze the physiological, biochemical and molecular parameters of tree plants of the Paricá species (Schizolobium amazonicum Huber ex Ducke) subjected to stress through CdCl2 measurements in the soil. For biometric analysis will be evaluated stem length, root, shoot and dry matter. On the biochemical relationship between proline glycine and Cd accumulation factor. In order to understand the molecular parameters, the RT-qPCR (Real Time Quantitative) technique will be used, using an endogenous gene (Actin-97) and 3 exogenous (HMA4, AtFC1_F and AQP TIP-1) that act in the mobilization of heavy metal in the plant, the amount of RNA will be measured by the UV/Vis spectrophotometer Biodrop Duo model. Purity will be calculated through the ratio of absorbances measured at 260 and 280 nm (ratio 260/280) and results close to 1.8 will be considered as having an acceptable degree of purity. The experiment will be carried out in the greenhouse of the Instituto de Ciências Agrárias of the Universidade Federal Rural da Amazônia-UFRA, in Belém-Pa, with geographic coordinates of 01º 27' 21” S, 48º 30' 16” W and an average altitude of 10 m. . The experimental design will be completely randomized (DIC), with 4 treatments divided into Control CdCl2 0mg/L, CdCl2 10 mg/L, CdCl2 20 mg/L and CdCl2 30 mg/L with 6 replications each (one plant/pot), totaling 24 experimental units. Cadmium will be defined based on the guiding values of CONAMA nº 460/2013, for soils contaminated by cadmium. The data obtained will be submitted to statistical analysis, using analysis of variance (ANOVA) in Sisvar version 5.4 and the means compared by Tukey's test at 5% probability.
