Bartonellosis is a zoonotic disease caused by intracellular gram-negative bacilli of the genus Bartonella, capable of causing disease in dogs and cats, and has several species considered zoonotic, with B. hensalae being the main species responsible for causing disease in humans, even leading to death. Research related to the molecular evaluation of this bacterium in animal reservoirs can contribute significantly to its epidemiological characterization, genetic variability and identification of possible hosts. In this context, the present study aims to perform the molecular study of the occurrence of Bartonella spp. infection in domesticated cats in the metropolitan region of Macapá, State of Amapá. A total of 303 blood samples collected in 5 veterinary clinics in the study region were analyzed. Total DNA extraction was performed using a commercial kit. PCR assays were developed for partial amplification of the gltA gene of the genus Bartonella spp. (350 bp). The amplicons of the positive samples were purified and sequenced in the forward and reverse directions. The consensus sequences were obtained using the BioEdit software (v7.2.6) and submitted to the BLAST platform. They were then aligned with a database containing representative sequences of the 45 species of the genus available in the Genbank database. For phylogenetic analysis, the Maximum-Likelihood method and the Kimura 2-parameter model were used in the MEGA software v7.0 for taxonomic identification of the isolates. The nucleotide and amino acid distances were calculated using the Geneious software v8.1.3. The results of the samples analyzed using the traditional PCR technique obtained positivity in 20 samples (6.6%), of which four samples (1.32%) were sequenced and tested positive for the genus Bartonella spp.
