The objective of this study was to evaluate the effect of the incorporation of açaí extract into semen cryopreservation diluent on the morphology, kinetics and integrity of the plasma membrane of sperm cells. The ejaculates, obtained from five bulls with semen of low freezing performance, were fractionated and distributed according to the experimental group. The control ejaculate did not receive açaí extract, and for the others, 5, 10, 15 or 20 mg/mL of açaí extract was added to the diluent. Sperm morphology was evaluated with formalin-saline-buffered solution. The structural and functional integrity of the plasma membrane was evaluated by the supravital, hyposmotic and epifluorescent tests and the cellular kinetics by the computerized analysis of the spermatic movement. Sperm alterations presented a linear decreasing effect (P<0.05) with the addition of different concentrations of açaí extract. No influence (P>0.05) of the different concentrations of açaí extract on the structural and functional integrity of the sperm plasma membrane evaluated by the supravital and hyposmotic tests was observed. When evaluated by epifluorescence, the integrity of the plasma membrane was higher (P< 0.05) after the addition of acai extract to the cryopreservation diluent. There was no significant effect (P>0.05) of the addition of açaí extract on the kinetics of spermatozoa. The results allow suggesting the use of epifluorescence as a more efficient method to evaluate the integrity of the plasma membrane of spermatozoa. In addition, it can be concluded that the addition of acai extract to the freezing diluent guarantees better preservation of the structural integrity of the sperm plasma membrane in the semen with low tolerance to the cryopreservation process.
