SUMMARY:

Buffaloes play a notable economic, social and environmental role in the world. Milk is the second most produced in the world and has excellent levels of proteins, lipids, vitamins and minerals, and meat has excellent organoleptic characteristics. They are animals capable of adapting to the most diverse environments, transforming grasses into products with high added value, in addition to high-value waste, making them an interesting link in natural production systems. However, enhancing milk production is not only used with good health, nutritional or environmental management conditions, it is necessary to understand and know the genetic factors at the level of DNA and RNA and how, combined with good management, they can influence the productivity of dairy products. animals. Thus, this research aimed to investigate single nucleotide polymorphisms (SNPs) in lipid and protein metabolism genes in order to associate them with data on milk production and quality in a herd of crossbred buffaloes from the Amazon region. For this, blood samples were collected from 85 buffaloes belonging to a property located in the municipality of Bujaru/Pa. Next, DNA extractions were carried out using the phenolic method from all samples. Temperature gradient tests were performed for the OLR1 and LALBA genes, which were standardized. Subsequently, conventional PCRs were performed on two samples (24 and 26) for both genes, then they were purified using a commercial kit following its recommendations and finally sequenced using the Sanger method. For the OLR1 gene, it was not possible to observe the presence of possible SNPs, probably due to the low number of sequenced samples. However, for the LALBA gene, even with two sequenced samples, it was possible to notice differences between the reference sequence deposited in GenBank and the two samples that were sent, these differences confer 6 possible SNPs, four of which are of the transition type at positions - 42 AG, -292 GA, -445 TC and -591 CT and two transversion types at positions -444 TG and -501 TG, in addition to a four-base indel (TAAA) at positions -552 , -553, -554 and -555. Samples of both genes were edited and aligned in Finch TV v 1.4.0 and BioEdit 7.2 software using ClustalW multiple alignment and compared with the reference sequence using BLAST. Therefore, based on this information, analyzes will be carried out on the remaining animals to continue detecting other SNPs, as well as proximate analysis and milk somatic cell count (SCC), gene expression profile and association of these data with zootechnical indices of milk production.
