SUMMARY:

High temperatures remove the organisms from their comfort zone, causing thermal stress and negative impacts to the cells. Thus, we aim to evaluate the expression of genes involved in the term tolerance (HSPA1A, HSPA2 and HSPA8), associating the expression of genes: pro-apoptotic (Bax), anti-apoptotic (Bcl2) and pro-inflammatory (IFN-γ) in primary culture of bovine and buffalo dermal fibroblasts submitted to thermal challenge, for 12 hours, in the following treatments: 0 (38.5 ° C / 24 hours); 3 (42 ° C / 3 hours); 6 (42 ° C / 6 hours) and 12 (42 ° C / 12 hours). The samples will be evaluated for cell viability, by the trypan blue test and gene expression by the qRT-PCR method. Relative gene expression will be determined by the 2^-ΔCt method, using GAPDH and β-actin as reference genes, the data will be tabulated in a spreadsheet and then tested for normality by the Kolmogorov-Smirnov test. A significance test will be applied through ANOVA according to the species effect, time effect and the interaction between species and time. The comparison between the averages will be through the SNK test. The effects of time, when significant, will be assessed through linear and quadratic regression analyzes, with a significance level of 0.05. All analyzes will be carried out using the SAS program (university version).
